How Does pH Buffer Affect Catalase’s Rate Of Reaction? - College Essay

How Does pH Buffer Affect Catalase’s Rate Of Reaction?

Factors that affect rate of reaction

Research Question:

How does pH buffer affect catalase's rate of reaction?

Introduction:

pH buffer:

a mixture of a weak acid and its conjugate base or a weak base and its conjugate acid. It is used to prevent changes in the pH of a solution. Buffer solutions are used as a means of keeping pH at a nearly constant value.

Hydrogen Peroxide: (H2O2)

Hydrogen peroxide is a clear colorless liquid, slightly more viscous than water. Due to its oxidizing properties, hydrogen peroxide is often used as a bleach or cleaning agent.

Catalase:

Catalase is an enzyme (catalyst) that catalyzes the decomposition of hydrogen peroxide to water and ...

pH instability will cause the change in the bonds in the enzyme, which change the enzymes composition and therefore cause its denature. As enzyme was functioned by the lock and key theory, substrate (key) must binds with specific enzyme's activation site (lock), which has a specific shape. Therefore the denature of the enzyme will make the reaction lose an alternative pathway to form product with an lower activation energy and thus lower the rate of reaction.

Variables:

Independent Variable:

pH of hydrogen peroxide (pH 3,5,7,10,13)

Dependent Variable:

Rate of reaction (amount of oxygen produced)

Reasons for Measuring

Ways to Measure

Because oxygen is the product of the reaction therefore measuring the change of oxygen concentration in air content could measure the rate of reaction.

Through oxygen gas sensor and gas chamber so that oxygen gas produced would not escape and can be sensed by the oxygen gas sensor.

Control ...
