Experiment outline

Research Question: how does ethanol effects on microbial growth?

Aim: to find out how ethanol effects on microbial growth.

Hypothesis: if the concentration of the ethanol is increasing, the more bacteria will by killed because the stronger ethanol, the more it can dissolve the lipid membrane of bacteria.

Variables:

Independent variable is the concentration of ethanol
Dependent variable is the amount of Bacteria
Controlled variable is the amount of ethanol and the amount of bacteria use

Making Agar plate:

Materials and equipment:
Schott bottle
Distilled water
0.6g Yeast extract (Borvil)
1.6g Sodium Chloride
3g Agar
1g peptone

The melting point of agar is around 600C, so it will be very warm when solidifying, so do not wait until it is cool!
Remove sterile petri dishes from plastic bag (take only what you need and re-sealing it if there are some remaining petri-dishes)
Working aseptically, pass (do not hold in the flame!) the neck of the bottle through the flame
Pour a thin layer of liquid Agar into each plate to only just cover the bottom of the petri-dish. Ensure that the lid is not lifted off excessively (see diagram below-you should open it up just enough to pour)
Swirl plate gently in a circular motion to ensure an even distribution of agar on the plate. NOTE: do not allow the agar to move up the side of the plate as this often lead to contamination.
Let each plate cool until it is solidified (20 minutes) then invert so as to avoid condensation settling onto the agar
Stack in 6 plats with cello tape
Label the cellotape with your group name, class and date. Store plates inverted ...
